Sample Submission Requirements
Bacterial liquid
Please offer the above 200μl fresh bacterial liquid, keeping in 1.5ml microfuge tube and sealing by sealing membrane, when we receive it, we will culture the bacteria free of charge, and extract the plasmid DNA from bacterial liquid. LB medium usually for us, culture temperature is 37¡æ, ampicillin, kanamycin, chloramphenicol are conventional antibiotics available. Please provide 4ml fresh bacterial liquid for us if you need special culture conditions , we can extract plasmid DNA directly from bacterial liquid you offered; please offer your store solution and its concentration to us if you used special antibiotics. In addition, we are glad to receive the plate bacteria, puncture bacteria, glycerol bacteria.
Plasmid
You can provide your plasmid for us if it meets the requirements of direct sequencing, OD260/OD280=1.8~2.0, plasmid concentration >200ng/μl, volume>10μl, but you are not encouraged to provide your plasmid.
DNA template
Please provide purified DNA to us.
Forms: Phage DNA; Low copy plasmid; Cosmid, BAC DNA.
Dissolved purified DNA in sterile deionized water, DNA concentration>200ng/μl, (DNA concentration >500ng/μl if vector is length), volume>10μl, we will detect the DNA template you offered. You need to show the name and plasmid profile, the range of insert fragment and enzyme sites, name and sequence of sequencing primers, the distance between sequencing primers and insertion site. Sequencing primers you offered should be purified by PAGE, primers concentration>5pmol/μl.
PCR products
If you offer the PCR products, we detect by agarose gel firstly, when the band is clear, we recover and purify the target band on gel. We will charge for the purification service whatever the sequencing result is. If you provide the purified PCR products, you had better to dissolve it in sterile deionized water (Concentration> 50ng/μl, volume>10μl,), while providing the PAGE purified primers used for PCR (primer concentration> 5pmol/μl).
Insertion fragment
Please offer purified cosmid DNA and show its concentration, DNA quantity>5μg. You should also provide primers for us, primers volume>10μl, primers concentration>5pmol/μl.
Note
When you send the orders to us, please provide the information of all the required items carefully. Such as, primers name, numbers of reaction, sequencing in one direction or bi-direction etc.
